Nanobiotechnology: Versatile 3-D nanostructures using DNA 'bricks'

ScienceDaily (Nov. 29, 2012) ? Researchers at the Wyss Institute for Biologically Inspired Engineering at Harvard University have created more than 100 three-dimensional (3D) nanostructures using DNA building blocks that function like Lego? bricks -- a major advance from the two-dimensional (2D) structures the same team built a few months ago.

In effect, the advance means researchers just went from being able to build a flat wall of Legos?, to building a house. The new method, featured as a cover research article in the 30 November issue of Science, is the next step toward using DNA nanotechnologies for more sophisticated applications than ever possible before, such as "smart" medical devices that target drugs selectively to disease sites, programmable imaging probes, templates for precisely arranging inorganic materials in the manufacturing of next generation computer circuits, and more.

The nanofabrication technique, called "DNA-brick self-assembly," uses short, synthetic strands of DNA that work like interlocking Lego? bricks. It capitalizes on the ability to program DNA to form into predesigned shapes thanks to the underlying "recipe" of DNA base pairs: A (adenosine) only binds to T (thymine) and C (cytosine) only binds to G (guanine).

Earlier this year, the Wyss team reported in Nature how they could create a collection of 2D shapes by stacking one DNA brick (42 bases in length) upon another.

But there's a "twist" in the new method required to build in 3D.

The trick is to start with an even smaller DNA brick (32 bases in length), which changes the orientation of every matched-up pair of bricks to a 90 degree angle -- giving every two Legos? a 3D shape. In this way, the team can use these bricks to build "out" in addition to "up," and eventually form 3D structures, such as a 25-nanometer solid cube containing hundreds of bricks. The cube becomes a "master" DNA "molecular canvas"; in this case, the canvas was composed of 1000 so-called "voxels," which correspond to eight base-pairs and measure about 2.5 nanometers in size -- meaning this is architecture at its tiniest.

The master canvas is where the modularity comes in: by simply selecting subsets of specific DNA bricks from the large cubic structure, the team built 102 3D structures with sophisticated surface features, as well as intricate interior cavities and tunnels. "This is a simple, versatile and robust method," says Peng Yin, Ph.D., Wyss core faculty member and senior author on the study.

Another method used to build 3D structures, called DNA origami, is tougher to use to build complex shapes, Yin said, because it relies on a long "scaffold" strand of DNA that folds to interact with hundreds of shorter "staple" strands -- and each new shape requires a new scaffold routing strategy and hence new staples. In contrast, the DNA brick method does not use any scaffold strand and therefore has a modular architecture; each brick can be added or removed independently.

"We are moving at lightning speed in our ability to devise ever more powerful ways to use biocompatible DNA molecules as structural building blocks for nanotechnology, which could have great value for medicine as well as non-medical applications," says Wyss Institute Founding Director Don Ingber, M.D., Ph.D.

The research team led by Yin, who is also an assistant professor of systems biology at Harvard Medical School (HMS), included Wyss Postdoctoral Fellow Yonggang Ke, Ph.D., and Wyss Graduate Student Luvena Ong. Another contributor was Wyss Core Faculty member William Shih, Ph.D., who also holds appointments at HMS and the Dana-Farber Cancer Institute.

The research was supported by the Office of Naval Research, the Army Research Office, the National Science Foundation, the National Institutes of Health, and the Wyss Institute for Biologically Inspired Engineering at Harvard University.

Insight: Cash crisis, Arab ferment threaten Jordan's stability

AMMAN (Reuters) - Violent protests that shocked Jordan this month have mostly subsided, but unprecedented chants for the "fall of the regime" suggested a deeper malaise in a kingdom so far spared the revolts reshaping the Arab world.

Anger over fuel subsidy cuts undoubtedly drove the unrest, in which police shot dead one man during a confrontation at a police station. The government's planned electricity price rises starting next year may well ignite more popular fury.

King Abdullah has made some constitutional reforms and his counselors say turnout at a parliamentary poll in January will test public support for the pace of political change amid an acute financial crisis that has forced Jordan to go to the IMF.

However, the model that has kept Jordan relatively stable for decades is cracking, nowhere more so than in the tribal East Bank provinces long seen as the bedrock of support for the Hashemite monarchy installed here by Britain in 1921.

The formula reinforced after the 1970 civil war between the army and Palestinian guerrillas - a defining national trauma now airbrushed from public discourse - broadly gives East Bankers jobs in the army, police, security services and bureaucracy.

Jordan's Palestinian-origin majority dominates private enterprise, but does not play a commensurate political role, in part because electoral gerrymandering curbs its voting power.

Although the fissure between the two communities is blurred by inter-marriage, long co-existence and, at least among the elite, business ties, it is likely to haunt Jordan as long as the wider Israeli-Palestinian conflict remains unresolved.

Jordanians of all stripes are fearful of the insecurity that stalks their neighbors, but the money that kept discontent in check across a fragmented society is simply no longer there.

An influx of 240,000 refugees from the Syrian conflict next door has further strained the resources of a country of seven million that has almost no oil and precious little water.

"Reform is genuinely difficult because you need to change the economic as well as the political rules," said a European diplomat. "In the past the tribes gave their support in return for jobs and money. Now that this is no longer affordable, they are shouting things like 'We won't pay for your corruption'."

Palestinians, while also hard hit by the austerity measures, have mostly laid low to avoid political flak.

DISGRUNTLED TRIBESMEN

In Kerak, a tribal hilltop town caught up in price protests earlier this month, morose shopkeepers await customers in the narrow market streets below the imposing Crusader citadel.

"Everyone who feels the pinch should go out in the street to express his views peacefully," said Hani Herzallah, 41, a barber with four children. He said he had joined the protests against fuel price rises that included a 54 percent increase in the cost of gas cylinders most Jordanians use for cooking and heating.

At a shop selling live chickens from wire cages, Tahseen al-Tanashat, 64, said he had just drawn his 200 dinar ($280) pension, but only had 50 dinars left after paying his bills.

Tanashat, on a state pension since he retired as a guard 31 years ago, said two of his three sons were soldiers. "I just want my 19-year-old still at home to get a job in the army."

For all their complaints, Kerak, 90 km (56 miles) south of Amman, has been lavished with state funds, thanks perhaps to powerful Majali and Tarawneh tribal figures who have occupied top positions in the government and military for decades.

An illuminated four-lane highway leads to the town of 65,000, passing a power station and an industrial zone that is far from bustling. Kerak boasts a major university, a new public hospital along with training colleges, and a palace of justice.

But jobs are scarce. A government hiring freeze is meant to alleviate the public sector pay and pension burden on a state treasury long reliant on aid from Gulf Arab and Western donors.

A U.S. diplomatic cable published by WikiLeaks said Jordan's "bloated civil service and military patronage system" soaked up 83 percent of the 2010 budget, despite planned spending cuts.

The economy has hit even stormier seas since then. Egypt's new rulers have sharply reduced cheap gas supplies to Jordan, which imports 97 percent of its energy and which has suddenly had to pay an extra $2.5 billion a year for fuel.

This month's protests were the most violent of several bouts of unrest in Jordan since Arab uprisings erupted nearly two years ago and toppled autocrats in Tunisia, Egypt and Libya.

Those in Kerak and other East Bank towns were organized by local opposition movements known collectively as Hirak, whose grievances focus on corruption, poor services and unemployment. They also resent privatization and other market reforms intended to reduce state spending - from which they benefit.

"Hirak is not driven by democracy, but by a sense of entitlement," said Mustafa Hamarneh, a social scientist running for parliament in the provincial town of Madaba. "It has not developed from spontaneous mobilization into a national political movement. It is parochial, with personalized demands."

EMBOLDENED ISLAMISTS

Jordan lacks credible political parties, with the exception of the Muslim Brotherhood's Islamic Action Front, whose power base is mostly, but not exclusively, urban and Palestinian. In some cities Islamists have developed tentative links with Hirak.

The Brotherhood, which has a track record of moderation since its Jordan branch was licensed in 1946, plans to boycott the January election, citing rules it says are meant to keep it from securing the biggest bloc in the 150-seat assembly.

The authorities accuse the Islamists, emboldened by Arab uprisings that led to election wins for their counterparts in Egypt and Tunisia, of fomenting unrest and of refusing to join a reform dialogue launched by King Abdullah in early 2011.

"Apparently the Muslim Brotherhood decided they stood to get more gains if they stayed in the streets," said a senior official source, speaking on condition of anonymity.

He acknowledged that the timing of the subsidy cuts, just as winter and an election were approaching, was far from ideal, but said there was no choice because Jordan risked "insolvency".

In return for a $2 billion standby arrangement agreed in August, the International Monetary Fund wants public sector reform and action on subsidies, including electricity tariffs.

Gulf donors such as Saudi Arabia, which rescued Jordan from an earlier crunch point with $1.4 billion a year ago, have held off from giving direct budget support so far this year, though Riyadh and Kuwait have sent $250 million each for projects.

Speculation about the reasons ranges from heavy spending by Gulf nations to stave off disaffection at home, concern about corruption in Jordan, and more pressing regional priorities - or even irritation that Amman had factored assumptions about Gulf aid into its IMF presentation without asking the donors first.

Saudi Arabia and Qatar may also want Jordan to be more active in the Syria crisis. "They would essentially like to see Jordan becoming the southern equivalent of Turkey in supporting the Syrian opposition," said Amman-based analyst Moin Rabbani.

"The Jordanians however ... prefer to play a less visible role and exercise it more covertly."

The survival of a vengeful Bashar al-Assad or a triumph for his Islamist-dominated foes would both pose dangers for Amman.

Jordan, valued by the West for its peace treaty with Israel and for its role as a stable buffer in a volatile region, still has an ambassador in Damascus, in line with its usual policy of walking a careful line between its more powerful neighbors.

TOP-DOWN REFORM

When Arab revolts began last year, the king, reigning since his father Hussein died in 1999, renewed a political reform drive opposed by conservatives which he had set aside to focus on economic liberalization aimed at expanding the middle class.

"The results remain disappointing," wrote Julien Barnes-Dacey in a paper for the European Council on Foreign Relations. "Despite changes to the constitution, few restrictions have been placed on the king's direct political authority."

King Abdullah, who has replaced his cabinet five times in the past two years, can still appoint and dismiss governments, although he has promised to consult parliament on choosing the next prime minister, who must then win a confidence vote.

"Parliament must become its own master and not get dissolved by the king in two words," said Wisam al-Majali, a Hirak activist in Kerak. "Now if even the best parliament digs deeper on corruption, it is dissolved the next day."

Another Kerak activist, Moaz al-Batoush, said an empowered parliament would obviate the need for street protests against "stupid" decisions that risked igniting revolutionary demands.

"Some people angered by the price rises reacted by calling for the downfall of the regime," he said, adding that this had never been a Hirak demand. "There is a crisis of confidence."

The official source defended the reforms, which include creation of an independent electoral commission, saying an overwhelming majority of Jordanians opposed removing powers from a monarch seen as a safeguard amid competing interests.

He said re-drawing electoral boundaries was not easy, given resistance from now over-represented East Bankers - Amman gets only a fifth of seats in parliament, despite being home to roughly half Jordan's population, many of them Palestinians.

The mood is sour among Palestinians in the Hussein refugee camp, now a scruffy built-up neighborhood of the capital.

"These price rises have slapped people in the face," said Abdul-Moneim Abu Aisha, 52, a butcher dragging on a cigarette as he sold small gobbets of meat in a tiny neon-lit shop.

In a market street where stalls piled high with vegetables jut out into the snarled traffic, people said only minor fuel price protests had occurred in the camp. Some voiced suspicion that even these were the work of outside provocateurs.

"The Palestinian camps will move only when the Jordanian tribal cities move and when the whole country rises up. If the camps rise up on their own they will be put down brutally," said a carpenter, who gave his name only as Abu Omar.

"We are targeted as Palestinians," he said, while having his hair cut. "The first thing they ask when you enter a police station is about your original hometown. But I'm a Jordanian who served in the army, and if anything happens to the country I will be the first to defend it, so why ask where I come from?"

With East Bankers and Palestinians alike feeling aggrieved, tensions might calm if the January election produced a new-look parliament and a government with the popular legitimacy to take tough decisions, but the electoral rules and the planned boycott of the vote by Islamists and others make this unlikely.

While the 50-year-old king seems confident his roadmap is the best route for a divided society, not everyone is so sure.

"Jordan needs an inclusive political reform to cope with the horrendous economic challenges," the European diplomat said.

"What we have is a baby step. The democratic deficit remains and has not been narrowed at a time when you need public confidence to deal with the challenges and the corruption."

I was supposed to be listening to the cicadas and the sound of a flowing creek when a Mitsubishi van rumbled across a small steel bridge just downstream. It was probably depositing campers at a nearby tent village, where kids were running around with their fishing poles and pink bed pillows. This was nature, Japan style. I was in Chichibu-Tama-Kai National Park, a 75-minute train ride northwest of Tokyo, with half a dozen other hikers out for a dose of shinrin-yoku, or forest bathing. The Japanese go crazy for this practice, which is standard preventive medicine here. It essentially involves hanging out in the woods. It?s not about wilderness; it?s about the nature-civilization hybrid the Japanese have cultivated for thousands of years. You stroll a little, maybe write a haiku, crack open a spicebush twig and inhale its woodsy, sassy scent.

?People come out from the city and literally shower in the greenery,? our guide Kunio explained. ?This way they are able to become relaxed.? To help us along, Kunio?a volunteer ranger?had us standing still on a hillside, facing the creek, with our arms at our sides. I glanced around. We looked like earthlings transfixed by the light of the beamship. Or extras in a magical-kingdom movie. Kunio could have been one of the seven dwarves. Elfin, with noticeably large ears, he told us to breathe in for a count of seven, hold for five, release. ?Concentrate on your belly,? he said.

?When I?m out here, I don?t think about things,? he said.

In an effort to benefit the Japanese and find nonextractive ways to use forests, which cover 67 percent of the country?s landmass, the government has funded about $4 million in forest-bathing research since 2004. It intends to designate a total of 100 Forest Therapy sites within 10 years. Visitors here are routinely hauled off to a cabin where rangers measure their blood pressure, part of an effort to provide ever more data to support the project.

LibreOffice (for Mac)


We reviewed LibreOffice in 2011, and, as it was then, it's still the best no-cost alternative to Microsoft Office that you can find for Windows, Linux, and OS X. In this review, I focus on LibreOffice for OS X, and the Mac-specific reasons why you might or might not want it. I think you're far better off paying for Microsoft Office if you use Windows, while, under Linux, LibreOffice is your only choice if you want a full-featured office suite.

What is LibreOffice?
Some technical details first. LibreOffice is an offshoot from the old OpenOffice.org application suite, now renamed Apache OpenOffice. Like any other office suite, LibreOffice gives you a word-processor (Writer), a spreadsheet app (Calc), presentations and graphics apps (Impress and Draw), plus database, mathematical, and other tools. LibreOffice is fuller-featured and more up-to-date than any other OpenOffice variant, but it still suffers from a clunky interface with a 1990s look and feel.

On the Mac, if you need to share documents with a Microsoft-Office-centric world, you'll want Microsoft Office for the Mac, the only Mac application suite that's fully compatible with Windows-based formats, and a powerful and elegant app by any standard. The best Mac-native alternative to Microsoft Office is Apple's iWork suite with its dazzling graphics and high level of Office compatibility, and its ability to share and sync documents to other Macs and iOS devices through iCloud, but Microsoft's suite is the obvious first choice if you share documents with Windows users.

Libre Means Free
So where does that leave LibreOffice? For one thing, LibreOffice costs nothing, it exports to the latest Microsoft Office formats in addition to its native ODT format, and it's the obvious first choice for any organization that requires the use of open formats and open-source software. Also, LibreOffice is one of the few apps running under OS X that give you a fighting chance of importing legacy documents created by old Windows and MS-DOS apps?and even documents created by ancient Mac apps that almost no other current Mac app can open. For example, LibreOffice opens documents created by any version of WordPerfect, including the DOS, Mac, and Windows versions. This matters because law firms and many government agencies still have thousands of documents in WordPerfect format, and these might otherwise get orphaned if the office switched to the Mac.

LibreOffice also opens Microsoft Works documents that Microsoft Office for Windows can't open, and its graphics module opens vector graphics in CorelDraw and Visio formats. An Office Alternative?
As a Microsoft Office alternative, LibreOffice ranks as fair-to-good, which is a lot better than nothing. It's harder to use, less flexible, less easily automated, and a lot worse-looking. It opens almost any recent Word file with fairly faithful formatting, and does a good job of opening Excel worksheets if they aren't exceptionally complicated and don't use Excel's latest graphic features. If all you need is the ability to edit and save uncomplicated Word files on the Mac, then OS X's built-in TextEdit may be all you need, just as Windows' built-in WordPad may be all you need to edit such files under Windows.

As a tool for importing and converting legacy documents, LibreOffice ranks as good, which is better than any rival on OS X, but it can't match older utilities like MacLinkPlus (formerly available from DataViz) which won't run under recent versions of OS X. LibreOffice won't solve all legacy-file problems, however. For example, it can't open files created by the ancient MS-DOS versions of Microsoft Word?but neither can the latest Mac version of Microsoft Word. I keep wishing that Microsoft's Mac Business Unit will create import filters for legacy formats such as WordPerfect and Word for MS-DOS, but I've been waiting a long time, and I'm not very hopeful. Meanwhile, for anyone who really needs to run legacy DOS or Windows apps on a Mac, there's always the option to buy an app like VMware Fusion or Parallels Workstation that makes it possible to run a Windows app on the OS X desktop.

Other OpenOffices
LibreOffice isn't the only OpenOffice offshoot available for the Mac. A slightly more Mac-native branch of OpenOffice called NeoOffice is available for download to anyone who donates $10 per year. NeoOffice lags behind LibreOffice in its feature set, and its import filters tend to lack features built into LibreOffice's filters; for example, LibreOffice can import non-western alphabets in WordPerfect documents, but NeoOffice can't. NeoOffice has one major advantage over LibreOffice: it's far more stable. Under OS X?though not under Windows or Linux?LibreOffice crashes a lot more often than any modern app should, often when opening a document or changing an option. NeoOffice tends to stay open when performing the same operation.

LibreOffice isn't for every Mac user, but for anyone who has to work with oddball formats and legacy documents, it's an essential tool. And you can't argue with the price.

Online stores have cashed in on the idea, offering mouthwatering discounts on everything from cars to clothes to tempt consumers online on Singles Day, turning it into a retail bonanza? and it works.?

This year, e-commerce sales on Nov. 11 reached $4.6 billion, according to an estimate by the Economic Herald, a specialist daily in Shandong province.

That may not look like much compared with ?Black Friday? spending in the US (which topped $11.4 billion last year), until you take a couple of things into account:?

First, average Chinese salaries are 10 times smaller than average American salaries.

Second, this is just online. A comparable day in America might be ?Cyber Monday,? coming up after the weekend. Last year online US shoppers shelled out $1.25 billion ? little more than a quarter of what their Chinese counterparts spent two weeks ago.

The Chinese government may not be keen on the political corners of the Internet, but it loves the commercial aspect; its current five year plan for the economy foresees a fourfold jump in e-commerce?from 2010 levels to $2.9 trillion by 2015.

China has the world?s largest online population, at 538 million, and has more online shoppers than anywhere else too: On Nov. 11, some 213 million people ? nearly half of all Chinese Internet users ? visited one of Alibaba?s two retail platforms. Alibaba, which runs the two biggest e-commerce sites in China, reported sales of $2.94 billion on Nov.11.

Ten million consumers ? more than the population of Greece ? clicked on an Alibaba site in the first minute of Nov. 11, in the dead of night.

The massive Singles Day sales promotions are expected to boost the number of online shoppers even further. Ms. Liu for example, says she has normally shopped in bricks-and-mortar stores, but her Singles Day experience has converted her.

Love Sick Relationships ? Shio the Shiba

The most amazing thing about dog ownership is the relationship that you have with your dogs based purely on interpretation of each other?s behaviors. Shio understands my quirky habits better than I do and I always make the effort to figure out what he is trying to tell me. For example, if there is food on the counter he?ll sit on the floor closest to the bowl and stare at me until he grabs my attention. Then whips his head to look up towards the bowl, then look back at me. It?s very obvious what he?s trying to tell me there!

There are some less obvious interpretations that can only be known after co-existing for longer time periods. I came home early from work today to nap after feeling sick and found home greeting me at the door. He was not as enthused as normal but I was too groggy to realize. Then I went for a catnap in the bedroom and noticed that he climbed onto the bed to nap with me. Still groggy, I thought it to be a rare sign of empathy from our Shiba. I lightly napped and was awaken several times by his restlessness. Finally realizing that sneaking in a good nap was futile, I stood up and went back to my computer with him following closely behind. I had the inkling that something was wrong the whole time and finally followed my instincts and leashed him up for a walk.

Immediately upon walking out the door, Shio laid down on our welcome mat and vomited his breakfast. He then proceeded to quickly trot to our park to finish the rest of his business on a pile of leaves. This circle of excretion processes happened several times over the next 30 minutes and I took him immediately to the vet for some medication to ease his discomfort. I felt pain for having made him wait so long to go out and realized how considerate he was to hold it for as long as it could so that he wouldn?t soil the home.

He is now feeling much better and has been sleeping it off. Test results come back tomorrow.

Shio and I had a difficult year but moments like these make me realize how fragile we both are and how much I love him.

OECD urges gradual U.S. fiscal tightening to preserve growth

WASHINGTON (Reuters) - Tighter fiscal policy in the United States should be implemented gradually, the Organization for Economic Cooperation and Development said on Tuesday as it predicted moderate economic growth next year.

Automatic government spending cuts and tax increases, known as the "fiscal cliff", are set to begin in early 2013, draining about $600 billion from the economy unless the U.S. Congress and Obama administration agree on a plan to soften the blow.

In its latest economic outlook, the OECD said while cutting the large budget deficit was necessary to put the nation on a sustainable fiscal path, this should be done gradually and in the context of a well identified medium-term consolidation plan.

"The pace of consolidation should be gradual so as not to derail the already weak recovery," the OECD said. "If unresolved, the fiscal cliff would generate a significant drop in activity in 2013."

The United States has run budget deficits topping $1 trillion for three straight years, and it is on course to do so for a fourth.

The Paris-based OECD forecast growth in the world's largest economy of 2.2 percent from the fourth quarter of this year to the fourth quarter of next year, up from a projected 1.8 percent this year. Output is seen expanding 3.2 percent in 2014.

Annual gross domestic product expanded by an average of 2.1 percent over the last two years. The OECD forecasts assume the reduction in the U.S. budget deficit is limited to 1.5 percent of GDP in both 2013 and 2014.

But it said economic headwinds will continue to blow from the debt crisis in Europe and slowing global demand.

"The potential credit market disruptions associated with the ongoing crisis in the euro area remain a major concern," the OECD said. "Growth in disposable income will continue to be restrained significantly by the withdrawal of fiscal support."

OUTPUT GAP TO NARROW

But with U.S. economic growth expected to accelerate next year, the output gap -- the difference between the actual output and output at full capacity -- should narrow to 2.5 percent by the end of that year, the OECD predicted.

The output gap is currently 5.9 percent. Underpinning growth this year and in 2014 will be increased household spending, reflecting a turnaround in the housing market, which until recently was the economy's Achilles heel.

"Household balance sheets continue to improve, with equity values recently returning to near cyclical highs and home prices showing a sharp and sustained upturn," the OECD said.

"The gains in net wealth, coupled with low interest rates and a continued easing in lending conditions, should contribute to a pick-up in aggregate demand going forward."

It forecast a gradual improvement in the labor market and projected the unemployment rate falling to 7.5 percent over the course of 2014 from a projected 7.8 percent average in 2013.

The jobless rate was at 7.9 percent in October and almost all the decline from a peak of 10 percent in October 2009 was because of frustrated people giving up the search for work.

Given the still considerable slack in the economy, inflation pressures are likely to remain contained, even though wages could see rapid growth as the labor market improves, the OECD said.

This should help the Federal Reserve to honor its pledge to keep interest rates near record low levels through at least mid-2015.

The OECD forecast the consumer price index rising 1.8 percent over the course of next year and picking up to 2 percent over 2014.

The U.S. central bank, which has held overnight lending rates near zero since December 2008 and pumped about $2.3 trillion into the economy through bond purchases, aims for a 2 percent inflation rate.

"With significant slack in resource utilization and inflation expectations appearing well anchored, this highly accommodative stance of monetary policy is warranted," the OECD said.

Congo and M23 rebels negotiate in Uganda

KAMPALA, Uganda (AP) ? Congolese officials are in talks Sunday with representatives of M23, the rebel group that last week took control of the eastern Congo city of Goma, according to Ugandan officials.

Ugandan Defence Minister Crispus Kiyonga said that he is mediating discussions to help both sides reach a settlement that would end a violent rebellion that has sucked in Uganda and Rwanda, which both face charges of backing the rebels.

M23 President Jean-Marie Runiga is leading the rebels in the talks, according to Rene Abandi, M23's head of external relations.

Abandi, who is now based in the Ugandan capital Kampala, said M23 representatives met with Congolese President Joseph Kabila in a tense, two-hour meeting that was also attended by Ugandan President Yoweri Museveni.

"He tried to accuse us and we also tried to accuse him," Abandi said of the meeting with Kabila on Saturday. "It was a meeting to have a common understanding of the principle of negotiation. (Kabila) said he's ready to negotiate directly with us."

But some Congolese officials in the capital Kinshasa have said there will be no talks with the rebels unless they quit Goma. A regional summit of the International Conference on the Great Lakes Region in Kampala ? attended by both Kabila and Museveni ? on Saturday called on the rebels to leave Goma and urged Kabila to listen to the "legitimate grievances" of M23.

Despite the regional leaders' demands for the rebel forces to withdraw from Goma, M23 soldiers were visibly in control of the city Sunday. M23 also still held Sake, a contested town 25 kilometers (15 miles) west of Goma. The Congolese army attacked the town Saturday, but M23 retained control.

M23 President Runiga said that withdrawal from Goma was "under consideration" and, while M23 did not oppose the idea "in principle," no decision had been taken yet, according to M23 spokesman Lt. Col. Vianney Kazarama, speaking to the Associated Press. Runiga is still in Kampala and no official response to the demands from the regional summit is expected before his return to Congo, said Kazarama in Goma.

"We are waiting to hear from Runiga when he will be back from Kampala," said Kazarama.

"Since May we have asked to meet with President Kabila," said Amani Kabasha, M23's deputy spokesman. "At least now there has been contact. The door is open for talks to find the durable peace that eastern Congo needs."

Government troops remain in Minova, 25 kilometers (15 miles) south of Sake, following a failed attack on M23 last Thursday. Unruly Congo army soldiers had looted residents for the third night running, according to a United Nations official in the town who insisted upon anonymity because he is not authorized to speak to the press. U.N. peacekeepers patrolled Minova throughout the night to protect civilians from the rampaging government troops.

In Minova, Congo Gen. Francois Olenga, who was recently named head of the Congolese army, held meetings with area commanders . "The country is in danger. We cannot defend our country with traitors," said Olenga to The Associated Press.

Pickup trucks packed with Congolese army soldiers armed with automatic rifles and rocket propelled grenades sped through Minova to regroup at the local soccer stadium. Army soldiers were also walking in the streets, looking for food. Some army soldiers were selling cigarettes on the side of the road.

An M23 communiqu? sent Saturday night claimed that government regiments were moving into attack positions around rebel-held territory.

"Let them attack us!" said M23 spokesman Kazarama. "Do they have the strength? Absolutely not, we are in a strong position."

BEIRUT (AP) ? Syrian rebels on Monday captured a hydroelectric dam on the Euphrates River in the country's north after days of heavy clashes, carting off boxes of ammunition from defeated regime forces in the latest in a string of recent strategic advances for opposition fighters, activists said.

Also Monday, activists said rebels and pro-government Kurdish gunmen struck a truce to end days of fighting in the town of Ras al-Ayn near the border with Turkey that opposition forces entered earlier this month.

The Britain-based Syrian Observatory for Human Rights said rebel fighters overran regime defenses and captured the Tishrin Dam, near the town of Manbij, before dawn Monday. Observatory director Rami Abdul-Rahman said the dam supplies several areas of Syria with electricity.

"This is a major blow to the regime," said Abdul-Rahman by telephone, describing the dam as "strategic location" on the Euphrates, which flows from Turkey through Syria and into Iraq.

The rebels have scored a series of hard-fought strategic advances recently, perhaps even seizing the momentum in their relentless battle of topple President Bashar Assad's regime. On Sunday, they captured a regime helicopter base outside Damascus before pulling back for fear of government airstrikes.

Amateur videos posted online showed gunmen inside the dam's operations room as an employee sat in front of five screens speaking by telephone about the level of water behind the dam. Another video showed gunman in front of dozens of green wooden boxes apparently full of munitions.

A gunman opened one of the boxes showing that it contained hand grenades. "The Free Syrian Army has fully liberated the Tishrin Dam," one of the rebels could be heard saying.

The activist videos appeared genuine and corresponded to other AP reporting about the events depicted. Syria restricts the access of reporters.

Syria's conflict started in March 2011 as an uprising against Assad's regime, but quickly morphed into a civil war that has since killed more than 40,000 people, according to activists. It has also prompted hundreds of thousands of Syrians to flee to neighboring Lebanon, Jordan, Iraq and Turkey.

Turkey's state-run Anadolu Agency said airstrikes by regime warplanes killed and wounded several people in the Syrian border town Atmeh on Monday. The agency said some of the wounded were brought to Turkey for treatment, and several people were also heading to the Turkish border to seek refuge. Atmeh is about 130 kilometers (80 miles) west of the Tishrin Dam.

Because it is close to the border, Atmeh has emerged as a key smuggling route for aid and arms to rebel areas in the northern provinces of Idlib and Aleppo and as a base for many rebel groups fighting further south. Residents assumed that their proximity to the Turkish border kept them safe from airstrikes.

"This is the first time the jets have hit us here, but it's probably not the last," said Musab Kadour, an Atmeh resident.

A local official in the Turkish town of Reyhanli confirmed that Turkish F-16 jets flew over the border region Monday. He spoke on condition of anonymity because he was not authorized to speak to reporters.

The Observatory reported air raids on other areas as well, including the northern towns of Maaret al-Numan, Kfar Rouma and Harem.

Also Monday, Kurdish activist Mustafa Osso and the Observatory's Abdul-Rahman said the town of Ras al-Ayn has been quiet since Saturday after fighters from the government-leaning Kurdish Democratic Union Party, or PYD, reached an agreement on a truce with rebels, most of them from Islamist extremist groups, including the al-Qaida-inspired Jabhat al-Nusra.

The rebels entered the town earlier this month and have since clashed almost daily with PYD gunmen for control over the area. Both factions add to the complexity of Syria's conflict, which has taken on heavy sectarian ? and ethnic ? overtones.

The fighting in recent days has left dozens of people killed or wounded, and dozens captured on both sides. Osso and Abdul-Rahman said PYD forces and the rebels agreed to exchange prisoners and to withdraw their militiamen from the town.

Abdul-Rahman said the rival parties agreed to form a local council that will run day to day life in Ras al-Ayn. Osso said thousands of people who fled the town, which has a mixed population including Arabs, Kurds, Chechens and Christians, have started returning home.

A journalist in Ras al-Ayn told The Associated Press that the town has been calm since Saturday, adding that cars with PYD gunmen were seen in the streets with loudspeakers urging residents to return to their homes after the truce was reached. He added by telephone that parts of the town were heavily damaged by government air raids earlier this month.

When regime forces withdrew from Kurdish areas in northeastern Syria in July, they were quickly replaced by Kurdish fighters from PYD. Those forces then battled rebel fighters after they pushed their way into predominantly Kurdish areas. The Kurdish group is affiliated with the PKK, rebels fighting for autonomy in the Kurdish-dominated southeast region of Turkey.

The Islamic militants, who are fighting on the side of the rebels, have played a bigger role in the Syrian conflict in recent months and many openly say they want to set up an Islamic state. The opposition is split, with some groups strongly opposed to the influence of extremists.

The Observatory also said that the health of leading opposition figure Abdul-Aziz al-Kheir, who it said is being held by one of the country's security agencies, has deteriorated recently. It did not disclose details of his health problems.

The National Coordination Body for Democratic Change in Syria accused the regime of being behind the disappearance of two of its leaders, including al-Kheir, shortly after they arrived home from China in September.

Al-Kheir, 61, comes from Assad's hometown of Kardaha and belongs to the president's ruling Alawite minority sect. He spent long periods in jail in the past because of his opposition to the regime.

Powerball jackpot builds to $325M for Saturday

(AP) ? The jackpot for Powerball's Saturday drawing has climbed to $325 million, the fourth-largest in the game's history.

A single winner choosing a cash payout would get nearly $213 million before paying state and federal taxes.

Powerball jackpots have been growing faster since the game's organizers doubled the ticket price to $2 in January.

An Iowa couple won $202 million in a Sept. 26 drawing, and then an anonymous winner in Delaware got $50 million prize the next week. The current jackpot has been building since then.

Powerball, which began in 1992, is played in 42 states, the District of Columbia, and the Virgin Islands.

January is often seen as a slow month, marked by post-holiday sales, credit card debt, and new fitness regimes. But for Chinese communities (both at home and abroad) it?s the run-up to the New Year festival, which falls on February 10th in 2013.? The weeks beforehand are some of the most important for retailers targeting this huge market. And the Chinese government? heavily encourages? shopping during the five-day holiday period.

Diwali, in November, is a huge celebration in India ? and increasingly the United States. The festival is a time for giving gifts, buying new clothes, decorating homes and streets, and holding firework displays. A growing number of Indians are now shopping online to avoid the crowds. Other big holidays include Eid, marked throughout the Muslim world.

For businesses who are already looking overseas, it?s worth considering other seasonal trends. For example, the ?back-to-school? season will vary from one country to another, and can be lucrative for retailers.

There are also marked trends in when people tend to take vacations ?which aren?t always easy to predict. The French and Italians tend to take much of? August off, while it?s more likely to be July in? Sweden. Many other Europeans prefer taking a winter vacation. And of course, December to February is the peak season in Australia and New Zealand.

Saturn meets Venus in dawn sky this week

For the last month or two, the planet Saturn has been on the far side of the sun, but emerges this week at dawn right next to the brilliant planet Venus.

Most of the planets appear to move from west to east against the background stars: right to left in the Northern Hemisphere and left to right in the Southern Hemisphere. Things are a little bit different when we are referring to rising planets in the dawn sky, relative to the point of sunrise.

In this case, both Venus and Saturn are on the far side of the sun, viewed from the moving planet Earth. The result is that Saturn appears to be moving away from the sun while Venus appears to be moving toward the sun. The two planets will appear to pass on the morning of Nov. 27, when they will be separated by about the width of the moon.

Venus is close to the sun and close to us, which combine to make it very bright. Saturn is far from the sun and far from Earth, combining to make it appear very dim in contrast to brilliant Venus.

Venus is a bright but featureless white gibbous shape, 87-percent illuminated, like a miniature version of the moon when it is four days past full, while Saturn is girdled by its famous rings.

Although Venus is much closer to us than Saturn, 1.4 astronomical units away (130 million miles, or 209 million kilometers), as compared to Saturn at 10.6 astronomical units (985 million miles, or 1.5 billion km), the two appear almost the same size in the telescope, because Saturn is a much larger planet than Venus.

This juxtaposition of Saturn and Venus in the sky is what astronomers call a conjunction, a fairly common event. It also forms a three-object alignment: Saturn, Venus, and Earth fall very close to a straight line.

There will be an even closer alignment of three solar system bodies on Dec. 21 this year. This is not the alignment you may have heard of associated with the 2012 Mayan apocalypse hoax.

On Dec. 21, there will be a transit of Venus visible only from the planet Saturn. Like the transit of Venus visible from Earth earlier this year, the planet Venus will appear to cross the face of he sun, but you would have to be near Saturn to see it. This is another three-object alignment: sun, Venus, and Saturn.

There are, of course, no humans anywhere near Saturn, but there is a sophisticated probe named Cassini in orbit around Saturn, and astronomers plan to use Cassini to observe this transit.

As to that other "planetary alignment" predicted for Dec. 21, supposedly involving all eight planets, that is quite simply impossible. In fact, all eight planets have never aligned, not even once, in the entire 4 billion plus years that the solar system has existed.

Because the changing positions of Saturn and Venus will make an interesting dance in the predawn sky, plan on observing a few mornings before and after the date of closest approach on Nov. 27. This is an excellent photo opportunity, and we will welcome your pictures of this event.
